My Project: Currently exploring how adipogenesis can be stimulated and inhibited by specific reagents.

Experiment of 12-October-2006

The experiment was designed to test qualitatively the difference in adipogenesis among 3T3 cells when different inhibitors, or stimulators were added. This experiment used a 6-well plate to culture the cells. The standard used was composed of .5uM of insulin, 1uM of Dexamethasone, and 1uM of Iso-Butyl-Methylxanthine. 2mL of the IDI standard was added to each well. Protocol consisted of splitting the cells and waiting 5 days to confluency, then adding the IDI standard and inhibitors and waiting 2 days. After the 2 days, just an Insulin standard was used along with the inhibitors for 5 days. After the 5 days, the same medium as before, an Insulin standard and the inhibitors, was used for 2 more days. On the last day, the cells were fixed, and stained with Oil Red-O. The overall process from splitting to staining took 14 days.

Well #1

This well contained only the IDI standard. As seen by the photograph, solely the IDI standard was able to cause minor differentiation of the fibroblasts.

Well #2

This well contained the IDI standard and 2uM of Rosiglitazone. As can be seen by the photograph, Rosiglitazone seemed to promote lipid accumulation, but slightly hinder cell proliferation. This is clearly evident from the lack of cells in spaces, but also the enlarged nature of the adipocytes.

Well #3

This well contained the IDI standard and 2uM of Indomethacin. Relative to Rosiglitazone, Indomethacin seemed to cause more proliferation in the cell, while decreasing lipid uptake. While there is still much lipid in the cells, there are more cells present with Indomethacin than Rosiglitazone.

Well #4

This well contained the IDI standard and 2uM of Indomethacin along with 2uM of Rosiglitazone. Intuitively, adding both stimulators would seem to cause rapid proliferation and lipid uptake, however, this was not the case. When both were added, the results depicted a somewhat medium between the previous results of each inhibitor by itself. Proliferation was better than with solely Rosiglitazone, however, lipid uptake was better than with solely Indomethacin.
